Abstract: THE ACTION OF OREXIN B ON PASSIVE AVOIDANCE LEARNING.ROLE OF TRANSMITTERS. G.Telegdy, A.Ekwerike. Dept.Pathophysiology,Medical School,Szeged. The action of orexin was studied on passive avoidance learning in rats Orexin B facilitated learning,consolidation of memory and retrieval of memory in a one trial step-through ,passive avoidance paradigm in a dose- dependent manner.Orexin B was administered into the lateral brain ventricle in conscious animals.The possible role of transmitters in mediating the action of orexin B was studied by pretreating the animals with receptor blocker with the dose proved to be effective in attenuating or blocking the action of the neuropeptides in the same paradigm.The following receptor blocker,in the dose used,could block the action of orexin B.:bicucullin.The following receptor blockers could attenuate but not fully block the action of orexinB:phenoxybenzamine,atropine,propranlol and naloxone,Haloperidol was ineffective.The data showed that orexin B can improve learning and memory processes following icv administration in a passive avoidance paradigm.In the action of orexin B,number of transmitter receptors such as GABA adrenergic,cholinergican dopiater receptors could be involved.The animals were kept and handled during the experiments in accordance with the instructions of the ethical committee of the University of Szeged for protection of animals. The effects of rat urocortin have been studied on learning,consolidation of learning and retrieval of memory in a one –trial step-through passive avoidance paradigm.The role of transmitters has been studied by pretreating the animals with different receptor antagonists,Urocortin given into the lateral brain ventricle facilitated the consolidation of memory in dose dependent manner.Selected dose of Urocortin could facilitate learning and retrieval of memory also.Pretreating the animals with haloperidol and atropine attenuated the action of urocortin on memory consolidation,however could not block the action completely.Nitro-arginin which blocks the action of nitric-oxide synthase enzyme thus blocking the nitric oxide formation,also attenuated the action of urocortin on consolidation of memory.The results demostrate that urocortin is able to facilitate the learning and memory processes in a passive avoidance paradigm.In this action,number of transmitters could be involved mediating the action of urocortin.
